Optimization of hairy root induction in the medicinal plant Bilhar (Dorema aucheri) using Agrobacterium rhizogenes for the production of secondary metabolites

Dorema aucheri, commonly known as Bilhar, is a perennial medicinal plant belonging to the Apiaceae family, known for its diverse and significant medicinal properties. The aim of this study is to investigate the induction of hairy roots in the medicinal plant Bilhar using Agrobacterium rhizogenes to optimize the production of secondary metabolites in in vitro conditions. This experiment was conducted factorially within a completely randomized design with three replications. The factors examined included explant type at three levels (hypocotyl, stem, and leaf) and bacterial strain at two levels (ATCC-15834 and A4). The results indicated that the interaction of the investigated factors significantly affected the traits of hairy root induction percentage, as well as the fresh and dry weights of the hairy roots after two months. The highest percentage of hairy root induction, along with the greatest fresh and dry weight, was achieved with the combination of the ATCC-15834 strain and hypocotyl explants, which was significantly higher than other treatment combinations. The transgenic nature of the hairy roots was confirmed by tracing a part of the rolB gene using PCR, while no corresponding band was amplified for natural (non-transgenic) roots. Overall, the findings of this study suggest that hairy root induction in Bilhar is influenced by both explant type and bacterial strain, and these results are an important prerequisite for experiments related to hairy root culture aimed at producing medicinal metabolites.
