fa شناسایی مولکولی گونه‌های Fusarium همراه چمن و برخی درختان در فضای سبز اردبیل Molecular identification of Fusarium species associated with turfgrass and some trees in Ardabil greenspace مهندسی ژنتیک میکروارگانیسم ها و ویروسها Microrganisms and Viruses پژوهشي Research <div style="text-align: justify;">در بین بیماری‌های مختلفی که گیاهان فضای سبز را تهدید می‌کنند، جنس Fusarium از اهمیت ویژه‌ای برخوردار است. با توجه به اینکه روش‌های ریخت‌شناختی در مورد Fusarium علاوه بر زمان‌بر بودن و نیاز به تخصص بالا، به‌تنهایی قادر به شناسایی دقیق و قابل اعتماد نیستند، استفاده از آغازگرهای اختصاصی روشی سریع، مقرون به‌صرفه و بدون نیاز به توالی‌یابی است که می‌تواند در شناسایی جنس و گونه Fusarium به‌راحتی مورد استفاده قرار گیرد. در این پژوهش، با توجه به‌ضرورت تایید مولکولی گونه های فوزاریوم همراه با زوال درختان زبان گنجشک، اقاقیا، صنوبر و زردی چمن در فضای سبز اردبیل، علاوه بر راست‌آزمایی دوباره آغازگرهای اختصاصی گونه با استفاده از جدایه‌های استاندارد، گونه جدایه های فوزاریومی به‌دست آمده از گیاهان فضای سبز با استفاده از آغازگرهای اختصاصی شناسایی شدند. در این پژوهش، تعداد 24 جدایه متعلق به گونه F. solani با استفاده از آغازگرهای fFuso1 وrfuso2 ، 11 جدایه متعلق به گونه F. oxysporum با استفاده از آغازگرهای CLOX1-F و CLOX2-R، 10 جدایه متعلق به گونه F. acuminatum با استفاده از آغازگرهای FAC-F و FAC-R، 8 جدایه متعلق به گونه F. equiseti با استفاده از آغازگرهای FEF1 و FER1 و 8 جدایه متعلق به گونه F. graminearum با استفاده از آغازگرهای FgCTPSf024 و FgCTPSrR306 شناسایی شدند. این آغازگرها هیچ واکنشی با جدایه های متعلق به سایر گونه های فوزاریوم مورد بررسی نشان ندادند. ضمناً از سه جدایه منتخب توالی‌یابی شده با آغازگر EF1-fus، دو جدایه به گونه F. solani و یک جدایه به گونه F. redolens تعلق داشتند.</div> <div style="text-align: justify;">Among the various diseases that threaten the green space plants, the Fusarium genus is of great importance. Due to time consuming, requiring high expertise and low reliability in the morphological methods, it would be recommended to use species-specific primers for identification of Fusarium species as a fast and cost-effective method without need to sequencing that could be easily used to identify the genus and species of Fusarium. In this study, in order to molecularly confirming of Fusarium isolates associated with Ash, Black locust and Populus decline, and turfgrass yellowing, in addition to verifying species-specific primers with standard isolates, Fusarium isolates obtained from Ardabil’s green space plants were identified using these specific primers by PCR. Totally, the 24 isolates belonging to F. solani using fFuso1 and rFuso2, 11 isolates belonging to F. oxysporum species using CLOX1-F and CLOX2-R primers, 10 isolates belonging to F. acuminatum species with using FAC-F and FAC-R primers, 8 isolates belonging to F. equiseti were identified using FEF1 and FER1 primers and 8 isolates belonging to F. graminearum were identified using FgCTPSf024 and FgCTPSrR306 primers. These primers did not react with isolates belonging to other Fusarium species.
