Investigation of use of tobacco rattles virus (TRV) to express the recombinant proteins in the tobacco plant

The object of this research was the investigation of efficiency of Tobacco rattle virus to express recombinant proteins in Nicotiana benthamiana. In the first step we produced a shorter version of the main vector. After addition of promotor and coding sequence for GFP protein to both vectors, they imported to two strains of Agrobacterium including LBA4404 and GV3101 and were inoculated leaves of 4 to 6 weeks' tobacco by them. At different times after inoculation, inoculated plants were photographed to determine the expression of GFP protein under UV irradiation. The parameters of the effect of type and size of vector, bacterial type and time effect on the percentage of leaf greens resulting from the expression of GFP protein were investigated. In 6-week-old plants that were injected with GPSpTRV2 (shortened) using LBA4404 strain was observed the maximum amount of green leaf area GFP, which is approximately 1.6 times the area of the green section was treated with the vector GPpTRV2 (not shortened) on the twentieth day after injection. In the 4-week-old plants fed with LBA4404 strain, the maximum amount of green areas in GPSpTRV2-containing plants was obtained on the 15th day, which is 1.4 times the maximum green area in GPpTRV2-containing plants in the same day. In 6-week-old plants, average area of green parts in the plants injected with both LBA4404 and GV3101 strains was approximately equal. Also, the level of plant leaf greens, which could indicate the expression of GFP, was time dependent and fluctuated in the days after inoculation.
