Analysis of P5CS1 Gene Expression Pattern in the Induction of Drought Stress Transcriptional Memory in
Arabidopsis thaliana

Plants frequently encounter repeated episodes of water deficit in their natural environment. Some plants can acquire an enhanced stress response upon re-exposure to the same stress, a phenomenon termed “stress memory”. This study investigates whether Pyrroline-5-carboxylate synthetase 1 (P5CS1), a key gene in proline biosynthesis, exhibits transcriptional memory in response to repeated dehydration stress in Arabidopsis thaliana. A transgenic line expressing the β-glucuronidase (GUS) reporter gene under the control of the P5CS1 promoter was subjected to two cycles of dehydration (4 h) and rehydration (4 h). Histochemical GUS staining revealed minimal expression after the first dehydration (S1), but strong expression after the first rehydration (R1) and significantly enhanced expression after the second dehydration (S2) and rehydration (R2). The proline content increased in each cycle; this increase was observed in the shoot after S2 and in the roots after R2 compared to the control. These results demonstrate that P5CS1 is a dehydration stress memory gene, with both its expression and proline accumulation being potentiated upon repeated exposure to water deficit. This work provides a functional link between proline metabolism and transcriptional memory in plant adaptation to recurring dehydration.
