fa شناسایی چندین توالی کدکننده پروتئین فسفولیپاز A2 در غده زهر عقرب همی‌اسکورپیوس‌لپتروس با استفاده از RNA-seq Identification of several coding sequences of phospholipase A2 protein in the venom gland of the scorpion of Hemiscorpius lepturus using RNA-sequencing مهندسی ژنتیک جانوری Animal پژوهشي Research <div style="text-align: justify;">عقرب Hemiscorpius lepturus ‌کشنده‌ترین عقرب ایران است و به همین جهت از نظر پزشکی اهمیت زیادی دارد. علائم بالینی ناشی از مسمومیت با زهر عقرب H. lepturus، مانند اثرات سمی نروتوکسیسیتی، سوزش‌های پوستی و همچنین برخی از واکنش‌های التهابی به فعالیت فسفولیپاز A2 (PLA2) آن نسبت داده شده است. هدف این مطالعه شناسایی و دسته بندی ژن‌های کدکننده فسفولیپاز A2 در غده زهر عقرب H. lepturus است. به همین منظور در این مطالعه، یک کتابخانه cDNA از غدد زهر این عقرب با استفاده از تکنیک توالی‌یابی RNA ایلومینا تولید شد. توالی‌های کد کننده PLA2 با استفاده از ابزار اصلی جستجوی تراز محلی (BLAST) و جستجوهای تشابه بین داده‌های خام غده زهر با توالی‌های ثبت شده از بندپایان دیگر به ویژه عقرب Centruroides sculpturatus شناسایی و استخراج شدند. این جستجوها منجر به یافتن چندین توالی همولوگ از PLA2 در غدد زهر این عقرب با شباهت توالی به فاکتور فعال کننده پلاکت استیل هیدرولاز (PAFA)، PLA2‌های وابسته به کلسیم (cPLA2)، PLA2‌های ترشح شونده (sPLA2)، PLA2‌های مستقل از کلسیم (iPLA2) و گروه III هترودیمریک فسفولیپاز A2 (hgPLA2) شد. اغلب توالی‌های یافت شده بیشترین شباهت را با توالی‌های PLA2 از عقرب‌های C. sculpturatus و Androctonus crassicauda نشان دادند. این گزارش یکی از معدود تلاش‌های توالی‌یابی رونوشت برای شناسایی، دسته بندی و تعیین خصوصیات فیزیکوشیمیایی توالی‌های PLA2 در غده زهر عقرب همی‌اسکورپیوس‌لپتروس است که در آن روابط فیلوژنتیکی بین بندپایان مختلف با استفاده از توالی‌های شناسایی شده از جنس Hemiscorpius  بررسی شده است.</div> <div style="text-align: justify;">The scorpion of Hemiscorpius lepturus is the deadliest scorpion in Iran, so it is very important from a medical point of view. Clinical symptoms caused by H. lepturus scorpion envenoming such as neurotoxic effects, skin burns, and some inflammatory reactions have been attributed to its phospholipase A2 activity. The aim of this study is to identify and categorize the genes encoding phospholipase A2 in H. lepturus scorpion venom gland. For this purpose, in this study, a cDNA library was produced from the venom glands of this scorpion using the Illumina RNA sequencing technique. The coding sequences of PLA2 were identified and extracted using the basic local alignment search tool (BLAST) and similarity searches between raw venom gland data with sequences recorded from other arthropods, especially the scorpion Centruroides sculpturatus. These searches led to the discovery of several homologs of PLA2 in the venom glands of this scorpion with sequence similarity to platelet acetylhydrolase-activating factor (PAFA), calcium-dependent PLA2s (cPLA2), secreted PLA2s (sPLA2), calcium-independent PLA2s (iPLA2) and, group III heterodimeric phospholipase A2 (hgPLA2). Most of the found sequences showed the most similarity with the PLA2 sequences from scorpions of C. sculpturatus and Androctonus crassicauda.
