Improving the growth of hairy roots in the medicinal plant Perilla (Perilla frutescens) through optimization of titanium dioxide (TiO₂) nanoparticles

The medicinal plant Perilla frutescens is of great importance due to its valuable secondary metabolites. However, the production of these compounds in nature is often limited. Hairy root cultivation is known as an efficient technology in plant biotechnology for the mass production of secondary metabolites. The aim of this study was to improve the growth and biomass of Perilla hairy roots using titanium dioxide (TiO₂) nanoparticle elicitor. In this regard, hairy roots were induced from leaf explants using Agrobacterium rhizogenes strain ATCC15834. Root transformation was confirmed by PCR amplification of the rolC gene. The transgenic roots were then treated with different concentrations of TiO₂ nanoparticles (0, 5, 15, and 25 mg/L) for two different times (24 and 48 hours). The results showed that the interaction effect of concentration and treatment time on the fresh weight of hairy roots was significant. The best treatment was a concentration of 15 mg/L TiO₂ nanoparticles with an immersion time of 48 hours, which produced the highest amount of biomass. Lower (5 mg/L) and higher (25 mg/L) concentrations did not have a significant stimulating effect. This study clearly shows that the optimal use of TiO₂ nanoparticles as an elicitor is an effective and promising strategy for scaling up the production of hairy roots of the medicinal plant Perilla in a tissue culture system.
