Green synthesis of ZnO and CuO nanoparticles using extract of moringa and Evaluation of their effects on Mo-CBP3 gene expression

The use of plant extracts in the phytosynthesis of nanoparticles is an environmentally friendly method which atural solvents are used.and, due to increased production and reduction of cost, time and energy, can be used as a suitable alternative to conventional methods such as physical and chemical methods.The purpose of this study was to biosynthesize zinc oxide nanoparticles using moringa plant extract and valuate the effect of their different concentrations (0, 100, 150, and 250 ppm) over two time period such as 48 and 72 hour on the relative expression of the Mo-CBP3 gene in Moringa plant. Structural analysis of ZnO and CuO nanoparticles using scanning electron microscopy and Energy Dispersive X-Ray Analysis showed that the shape of spherical particles and their dimensions were 25-37 and 32-36 nm, respectively. The results of analysis of variance showed that the application of CuO and ZnO nanoparticles caused a change in the  Mo-CBP3 gene expression pattern. The highest expression, 48 hours after treatment, was observed with just about 150 ppm of ZnO nanoparticles . By increasing ZnO nanoparticles concentration, a significant decrease occurred in the expression of the gene for 72 hours while treatment with 100 ppm of CuO nanoparticles in 48 hour after treatment has the most Mo-CBP3 gene expression. In the interaction treatment, it was observed that the combined application of copper and zinc oxide nanoparticles caused relative gene expression, so that during the application of concentrations of 150 ppm zinc oxide and 100 ppm copper oxide, the highest level of gene expression was observed 48 hours after the application of the treatment. Overall, the results of this study showed that  copper and zinc oxide nanoparticles elicitors had a positive effect on Mo-CBP3 gene expression and the highest expression was obtained  at 48 hours intervals.
