Production of Doubled Haploid Plants of Bell Pepper (Capsicum annuum) Through Anther Culture

| Anther culture is recognized as an efficient method for producing doubled haploid (DH) plants, serving as a powerful tool to accelerate plant breeding programs, especially in self-pollinated crops such as bell pepper (Capsicum annuum). In this study, four commercial F1 hybrids—Nirvin, Arancia, Tarento, and Massilia—were evaluated for their capacity to produce callus, embryos, and plantlets through anther culture. Flower buds at the appropriate microspore developmental stage (late uninucleate to early binucleate) were selected based on DAPI staining following a cold pre-treatment, and their anthers were cultured on five different media containing various concentrations of kinetin and 2,4-D. The experiment was conducted in a factorial arrangement based on a completely randomized design (CRD) with four replicates. Traits including callus induction, embryogenesis, and plant regeneration percentages were recorded. The results indicated that all treatments, except for the control (lacking plant growth regulators), induced callus formation. The highest callus induction rate (78.47%) was observed in the Massilia hybrid on medium C4, which contained the highest concentration of 2,4-D. In contrast, the highest embryogenesis rate (52.62%) was recorded in the Tarento hybrid on medium C2. However, plant regeneration was generally low, and only embryos derived from Arancia and Tarento successfully developed into complete plantlets. These findings highlight the critical role of both appropriate growth regulator combinations and genotype selection in the success of anther culture. Optimization of physiological, environmental, and culture conditions can significantly enhance DH plant production efficiency in bell pepper and contribute to breeding programs for this valuable crop |
