Comparison of the use of chemical and organic disinfectants for disinfection Syngonium podophyllum L. varieties under in vitro cultivation conditions

Syngonium podophyllum L. ornamental plant is widely grown as a houseplant due to its diversity and tolerance to low light conditions. Syngonium podophyllum is propagated by cuttings, but vegetative propagation on a large scale is time-consuming. In vitro culture provide for the production of more plants in a short period and the possibility of creating new varieties. Pathogens are considered to be the most important limiting factors in micropropagation. In addition to being unsuccessful, the use of conventional disinfectants may cause toxicity of the explants and in most cases cannot control all microorganisms. Attention has been paid to new alternatives to disinfectants. This study investigated disinfection with ethanol and household bleach, tobacco extract, cumin essential oils (20, 100 and 200 mg L-1), antibiotics (cefazolin, chloramphenicol, cefazolin, streptomycin and penicillin) and different application methods (in culture medium or immersion) of for Syngonium podophyllum cultivars in several experiments. The results showed that the in vitro contamination depends on the cultivar, method and type of disinfectant. Conventional disinfection methods were not effective in eliminating bacterial. The application of cumin in culture medium at low concentrations was able to prevent the growth of pathogens. Cefazolin 100 mg L-1was able to eliminate pathogens in some cultivars. Disinfection of explants with tobacco extract completely controlled in vitro contamination. According to the results, it can be recommended that by predicting the type of endogenous contamination of plants, compounds such as cumin and tobacco can be used for disinfection as a cost-effective, sustainable and environmentally friendly method.
