Development of high-throughput regeneration system from meristem apex culture of Cotton (Gossypium hirsutum L)

Cotton (Gossypium hirsutum. L) Is an economically important ‘cash crop’ widely used as a source of fiber and edible oil Development of a plant regeneration protocol for revitalize recalcitrant species like cotton (Gossypium hirsutum. L) from meristem apex culture with higher efficiency will put on a spurt the applications of functional genomics and selective breeding programs using transformation technologies. In present study we establish a highly efficient regeneration and micropropagation protocol of Iranian cotton cultivars. Among the tested combination, shooting medium (MS+ B5 vit+ 2 mg/l KIN+ 2 mg/l BAP+ 30 g/l glucose + 3g/l charcoal + 2.5 g/l phytagel) proved to be most suited for achieving the maximum number of multiple shoots..There was also no significant difference between cultivar’s shooting in percent. Finally shoots were transferred into five different rooting media. Statistical analysis of the results showed that the best medium for rooting is MS + 1mg/l IBA + B5 vitamins+ 30g-1glucose+ 2.5gl-1 phytagel+ 3g activated charcoal. The importance of present methods is shortening of regeneration time and improving the regeneration efficiency, as well as developing high quantity rooting protocol and induction genotype independent high number shoot per explant.
